Here at Oxford Expression Technologies Ltd, all the recombinant baculovirus services and projects are performed using our very own flashBAC™ baculovirus expression technology. This specialist technology has been modified to provide superior levels of expression for most protein targets. Like all Oxford Expression Technologies contract services, the baculovirus projects can cover every step from beginning to end.
Project support
With all Oxford Expression Technologies contract services, OET provides expert and comprehensive support from a number of experienced scientists from the beginning to end.
OET provides several its own proprietary transfer vectors (pOET series). These allow you to express genes under control of the very strong polyhedrin gene promoter or the p6.9 gene promoter. In choosing a transfer vector, there are several points to be considered; including the promoter, purification tag and signal peptide sequence.
OET has developed a rapid system based on quantitative PCR (qPCR) for the accurate titration of baculovirus stocks. In this method, the baculovirus DNA is purified from the budded virus using freshly amplified virus inoculum. the viral titre allows infection of the cells to be performed at a known multiplicity of infection (MOI) for maximum production of your recombinant protein.
Here at OET we have over 15 years experience in the production and purification of a wide range of proteins, which has been used for a combination of R&D projects, diagnostic work, vaccine development and more! For all our contract service work we tailor our methods to your requirements to ensure we maximise both the purity and yield using a wide range modifications.
Recombinant baculovirus is produced following co-transfection of insect cells with a transfer plasmid containing the gene of interest and flashBAC™ baculovirus DNA. The recombinant virus harvested from the co-transfection is then used to produce a high titre amplified virus stock.
To maximise yield, optimal multiplicity of infection (MOI), cell line and time of harvest is determined for optimal conditions for protein production.
Project completion
After completion of your project, we will provide a final report detailing the completed work for your project and we will provide help and assistance in shipping your plasmids, viruses, biomass and protein if required. We also offer a storage service which is useful for repeat and future work.
