OET can prepare up to 24 recombinant viruses simultaneously using a semi-automated robotic platform. In this case, we make an initial stock of 0.4 ml of each virus using a 24 well plate. To test expression, 100 ul of each virus is transferred to the wells of a fresh 24 well plate, seeded with Sf9 cells and 48 hours later, cell pellets and culture medium are tested for expression. Selected viruses can then be amplified to 50 ml as required for further study.
